Farnesyl Transferase Inhibitor
A-176120 is a selective inhibitor of farnesyl transferase with an IC50 of 1.2 nM. This compound demonstrates significant selectivity over geranylgeranyl transferase I (IC50=423 nM) and geranylgeranyl transferase II (IC50=3000 nM). A-176120 effectively inhibits Ras processing in H-ras transformed NIH3T3 cells and HCT116 K-ras mutant cells, with ED50 values of 1.6 and 0.5 μM, respectively. Additionally, A-176120 exhibits antiangiogenic and antitumor properties in vivo, significantly reducing capillary structure formation and vascular endothelial growth factor (VEGF) secretion.
