Antimitotic Agent

A-259745 is an orally active antimitotic agent that selectively targets the colchicine-binding site of tubulin. This compound demonstrates significant cytotoxic effects against both multi-drug-resistant and non-multi-drug-resistant cancer cell lines, exhibiting ED50 values of 0.018 µM for HCT-15 and 0.028 µM for NCI-H460. A-259745 inhibits tubulin polymerization, disrupts mitotic spindle dynamics, arrests cells in metaphase, and induces apoptosis. Its dose-dependent antitumor efficacy in murine models makes it a valuable tool for cancer research.
