ALR2 Inhibitor

ALR2-IN-9 is a potent inhibitor of aldose reductase (ALR2), exhibiting an IC50 of 21.8 nM. It demonstrates strong antioxidant properties, evidenced by an EC50 of 2.8 μM in DPPH radical scavenging assays. By directly interacting with reactive oxygen species (ROS) and reactive nitrogen species (RNS), ALR2-IN-9 disrupts free radical chain reactions and enhances the functions of catalase (CAT) and superoxide dismutase (SOD). Furthermore, it modulates the PI3K/Akt/Nrf2 signaling pathway to reduce hyperglycemia-induced mitochondrial superoxide production in vitro and alleviates oxidative stress in vivo. ALR2-IN-9 also extends the lifespan of C. elegans through the regulation of stress response genes, making it a valuable candidate for research into diabetic complications and anti-aging therapies.
