DAPK inhibitors target death-associated protein kinases, which couple stress signals to cell death. Death-associated protein kinase 1 (DAPK1) was cloned in 1995 from a screen for genes that mediate interferon-γ-induced cell death. DAPK1 is a calcium/calmodulin-regulated serine/threonine kinase with roles in apoptosis and autophagy. Its relatives include DAPK2, zipper-interacting protein kinase (ZIPK, also called DAPK3) and the DAPK-related apoptosis-inducing protein kinases (DRAK1, DRAK2). TC-DAPK6 and CK156 are listed as DAPK inhibitors, and HS38 acts on both DAPK1 and ZIPK. HS148 and HS94 are listed as DAPK3 inhibitors, while DRAK2-IN-1 targets DRAK2. PROTAC DAPK1 Degrader-1 is a proteolysis-targeting chimera (PROTAC) that induces DAPK1 degradation. DAPK Substrate Peptide serves as a phosphorylation substrate in kinase assays.
DAPK Substrate Peptide, synthetic peptide substrate for death associated protein kinase (DAPK) (Km = 9 uM).
PROTAC DAPK1 Degrader-1 selectively targets DAPK1 for degradation, exhibiting a DC50 value of 119.6 nM. This compound has been shown to elevate MDM2 protein levels while concurrently decreasing cleaved caspase-3 and cleaved PARP levels in a neurotoxin-induced cell apoptosis model. Its ability to effectively inhibit neuronal apoptosis positions PROTAC DAPK1 Degrader-1 as a valuable tool for investigating neurological disorders, including cerebral ischemia and traumatic brain injury.
DAPK1 ligand-1 is a PROTAC target protein ligand designed for the synthesis of PROTACs, including the DAPK1 Degrader-1. This compound facilitates the targeted degradation of LAG-3, demonstrating significant neuroprotective activity. Its application in research underscores its utility in studying the modulation of immune checkpoints and neuroprotection mechanisms in various biological contexts.

TAT-GluN2BCTM is a membrane-permeable peptide designed to target DAPK1 for degradation. By facilitating the transport of active DAPK1 to lysosomes, this compound effectively reduces its levels, thereby providing protection to neurons against oxidative stress and NMDAR-mediated excitotoxicity. TAT-GluN2BCTM is a valuable tool for research focused on neuroprotection mechanisms.
