RIP Kinase

RIP kinase inhibitors act on receptor-interacting protein kinases (RIPKs), mainly RIPK1, RIPK2 and RIPK3. RIPK1 and RIPK3 control necroptosis, a regulated form of necrosis triggered by tumor necrosis factor alpha (TNF-α) and other death receptor signals. Necrostatin-1 was described in 2005 as an inhibitor of nonapoptotic cell death, and RIPK1 was identified as its cellular target in 2008. A 2009 study showed that RIPK3 determines whether cells die by necrosis in response to TNF-α. GSK481 and PK68 are further RIPK1 inhibitors, and GSK-843 inhibits RIPK3. GSK583 inhibits RIPK2, which relays signals from the bacterial sensors nucleotide-binding oligomerization domain-containing proteins 1 and 2 (NOD1, NOD2).

    cRIPGBM chloride

cRIPGBM chloride is a selective modulator of receptor-interacting protein kinase 2 (RIPK2) that induces caspase 1-dependent apoptosis. This compound effectively disrupts the RIPK2/TAK1 prosurvival complex while promoting the formation of the RIPK2/caspase 1 pro-apoptotic complex. cRIPGBM chloride demonstrates significant anti-tumor efficacy in vivo in animal models, making it a valuable tool for research into glioblastoma multiforme (GBM) and related cancer therapies. Its proapoptotic properties offer insights into mechanisms of cell death and potential therapeutic strategies in cancer treatment.

    RIPK1-IN-31

RIPK1-IN-31 is a selective allosteric modulator of receptor-interacting protein kinase 1 (RIPK1) with an IC50 value of 16 nM. This compound demonstrates potential for modulating RIPK1 activity, making it a valuable tool for research in the fields of infectious, autoimmune, and neurodegenerative diseases. Its ability to selectively influence RIPK1 pathways can aid in elucidating the mechanisms underlying these conditions.
