BI-9627 hydrochloride

NHE1 Inhibitor

BI-9627 hydrochloride is a selective inhibitor of sodium-hydrogen exchanger isoform 1 (NHE1), exhibiting an IC50 of 6 and 31 nM in intracellular pH recovery and human platelet swelling assays, respectively. This compound demonstrates over 30-fold selectivity against NHE2 and negligible activity against NHE3 isoforms. BI-9627 hydrochloride effectively decreases autophagy in HTR-8/SVneo cells and can significantly lower the pH of human sperm, counteracting the effects of DMA. It further prolongs calcium recovery time in knockout hiPSC-derived cardiomyocytes and exhibits favorable pharmacokinetic properties along with low potential for drug-drug interactions, showcasing potent efficacy in models of ischemia-reperfusion injury.
