NHE Inhibitor
BMS-284640 is a synthetic inhibitor of the sodium hydrogen exchanger isoform-1 (NHE-1), demonstrating an IC50 value of 9 nM, which indicates over 380-fold greater potency than cariporide. This compound exhibits selectivity for NHE-1 over NHE-2 and NHE-5, with IC50 values of 1.8 μM and 3.36 μM, respectively. In addition to its potent inhibitory activity, BMS-284640 and its analogs show enhanced oral bioavailability and a prolonged plasma half-life in rat models, making it a valuable tool for research in cardiovascular and metabolic disease pathways.
