DI-1548

DCN1 Inhibitor

DI-1548 is a selective irreversible covalent inhibitor of DCN1, with an IC50 of 4.6 nM. It effectively inhibits cullin 3 neddylation while demonstrating minimal impact on the neddylation of other cullin family members. By covalently binding to DCN1, DI-1548 disrupts the DCN1-UBC12 interaction, leading to the collapse of the neddylation complex and subsequent inactivation of CRL3. This results in the accumulation of NRF2 and upregulation of its target genes, offering protective effects against liver damage. DI-1548 is suitable for applications in liver injury research.
