Sirtuins

Sirtuin inhibitors and activators target the seven mammalian sirtuins, a family of protein deacetylases. Yeast silent information regulator 2 (Sir2), the founding member, is a histone deacetylase that requires nicotinamide adenine dinucleotide (NAD+). Human sirtuins (SIRT1 to SIRT7) differ in location, with SIRT1 mainly nuclear, SIRT2 mainly cytoplasmic and SIRT3 to SIRT5 in mitochondria. Selisistat, also known as EX-527, inhibits SIRT1, and SirReal2 selectively inhibits SIRT2. SRT1720 HCl and SRT2104 are described as synthetic SIRT1 activators. Nicotinamide, a product of the sirtuin reaction, inhibits sirtuin activity, while nicotinamide riboside is a precursor of NAD+.
