CD73 inhibitors block ecto-5'-nucleotidase (CD73), the enzyme that forms extracellular adenosine. CD73 is a cell-surface protein held in the membrane by a glycolipid anchor. Together with ectonucleoside triphosphate diphosphohydrolase 1 (CD39), which breaks down ATP, it produces adenosine that suppresses T-cell responses, as shown for regulatory T cells. CD73 inhibitors are therefore used to study the adenosine pathway in tumor immunity and inflammation. Listed compounds include the nucleotide analogs AB-680 and MethADP, the inhibitors PSB-12379 and LY-3475070, and PD-L1/CD-73-IN-1, which also targets programmed death-ligand 1 (PD-L1).
