Kifunensine is a potent and selective inhibitor of class I α-mannosidases, originally isolated from *Actinomycete* species. It blocks the trimming of mannose residues from N-linked glycoproteins by inhibiting α-mannosidase I activity, thereby preventing proper glycoprotein maturation. Through this mechanism, Kifunensine disrupts endoplasmic reticulum–associated degradation (ERAD) pathways, leading to the accumulation of misfolded glycoproteins.
