L-699333 is a selective inhibitor of 5-lipoxygenase (5-LO), belonging to the thieno[2,3,4-cd]indole class. This compound effectively reduces the biosynthesis of 5-hydroxyeicosatetraenoic acid (5-HPETE) and leukotriene B4 (LTB4), with ICm values of 22 nM in human neutrophils and 3.8 pM in whole blood. L-699333 demonstrates anti-inflammatory and antiasthmatic properties across various animal models, including those of rat pleurisy, antigen-induced wheezing, and asthma in macaques and sheep. The compound exhibits high selectivity for 5-LO, showing less inhibition of human 15-LO and porcine 12-LO, positioning it as a valuable tool for investigating inflammatory pathways.
