NCI-006
LDH Inhibitor
NCI-006 is an orally active lactate dehydrogenase (LDH) inhibitor, with IC50 values of 0.06 μM for LDHA and 0.03 μM for LDHB. It effectively suppresses intratumoral LDH activity, lactate production, and tumor growth in mouse models of pancreatic cancer. NCI-006 inhibits glycolysis and promotes apoptosis in vitro, enhancing the radiosensitivity of glycolytic tumor cell lines while preserving normal cells. Additionally, it demonstrates synergistic antitumor effects when combined with IACS-010759 against colorectal and gastric cancers, and it impairs tumor growth in Ewing sarcoma models.
