OPN-9652

TEAD Inhibitor

OPN-9652 is a highly potent, orally bioavailable, covalent inhibitor of TEAD, exhibiting an IC50 of 0.005 µM in MSTO-211H cells. By targeting the central palmitate binding pocket of TEADs, OPN-9652 effectively diminishes TEAD-dependent reporter activity and downregulates the expression of key TEAD target genes, including CTGF and CYR61. This compound has been shown to resensitize drug-tolerant SOX10 knockout cells to combined BRAFi and MAPKi treatment, as well as to delay the onset of tumor resistance in a BRAF mutant A375 xenograft model. OPN-9652 is valuable for research in melanoma and related cancer pathways.
