TEAD Inhibitor

pan-TEAD-IN-1 is a potent pan-TEAD inhibitor that targets the palmitoylation site of TEAD, effectively disrupting its interaction with the coactivators YAP/TAZ. This disruption results in the inhibition of transcriptional upregulation of oncogenes such as Ctgf and Cyr61 within the Hippo signaling pathway. With a luciferase IC50 of 0.36 nM and an H226 cell IC50 of 1.52 nM, pan-TEAD-IN-1 demonstrates substantial biological activity. Additionally, it exhibits favorable pharmacokinetics and shows significant tumor growth inhibition in TEAD-dependent xenograft mouse models, indicating its potential utility in research involving TEAD-dependent cancers.
