PHGDH Inhibitor
PKUMDL-WQ-2201 is a non-NAD+-competing allosteric inhibitor of the enzyme phosphoglycerate dehydrogenase (PHGDH), exhibiting an IC50 of 35.7 μM. This compound effectively inhibits PHGDH mutant variants, demonstrating IC50 values of 69 μM for T59A and greater than 300 μM for T56AK57A. PKUMDL-WQ-2201 blocks de novo serine synthesis in cancer cells, thereby contributing to reduced tumor growth, making it a valuable tool for cancer research and therapeutic exploration.
