PolQi1
Polϴ Inhibitor
PolQi1 is a selective inhibitor of the Polθ domain of DNA polymerase, primarily targeting the Polθ-mediated microhomology end joining (TMEJ/alt-EJ) pathway. By inhibiting this pathway, PolQi1 significantly reduces insertion/deletion events and imprecise editing during DNA repair processes. This reagent enhances the efficiency and accuracy of homology-directed repair (HDR) and Prime editing, minimizing off-target effects. In combination with DNA-PK inhibitor AZD-7648, PolQi1 provides effective dual pathway regulation, making it a valuable tool for gene editing research, particularly in optimizing systems like CRISPR-Cas9 for challenging cell types such as primary hepatocytes and mouse embryos.
