Polθ Inhibitor

SY-589 is a potent inhibitor of the helicase domain of DNA polymerase theta (Polθ), with an IC50 of 2.29 nM. This compound disrupts Polθ-mediated microhomology-mediated end joining (MMEJ) DNA repair by inhibiting ATPase activity (IC50=0.85 nM) and promotes the formation of DNA double-strand breaks, as indicated by elevated γ-H2AX levels. SY-589 demonstrates significant antiproliferative effects in BRCA2-deficient cells, making it a valuable tool for research involving homologous recombination-deficient tumors.
