TDI-015051

Coronavirus Inhibitor

TDI-015051 is a selective, orally active antiviral agent that specifically inhibits the NSP14 guanine-N7 methyltransferase of coronaviruses. By binding non-competitively to substrates, TDI-015051 forms a stable ternary complex, effectively blocking the capping and methylation of viral mRNA, thus impairing viral replication and translation. It demonstrates potent antiviral activity against multiple coronaviruses, including SARS-CoV-2 and MERS, significantly reducing pulmonary viral load and inducing a moderate type I interferon-mediated immune response. TDI-015051 has IC50 values of 0.15 nM against SARS-CoV-2 and exhibits synergy with Nirmatrelvir while maintaining selectivity by not inhibiting non-coronavirus methyltransferases. This compound is suitable for research into COVID-19 and the replication mechanisms of coronaviruses.
