Thailanstatin A

RNA Splicing Inhibitor

Thailanstatin A is a potent inhibitor of eukaryotic RNA splicing, with an IC50 of 650 nM. It functions by non-covalently binding to the SF3b subunit of the U2 snRNA subcomplex within the spliceosome, displaying low-nM to sub-nM IC50 values across various cancer cell lines. Additionally, Thailanstatin A serves as an effective payload for antibody-drug conjugates (ADCs), including its conjugation to lysines on trastuzumab for the development of "linker-less" ADCs.
