Upacicalcet
Calcimimetic Agent
Upacicalcet is a non-peptide calcimimetic that acts as an agonist of the calcium-sensing receptor (CaSR) with an EC50 of 10.8 nM. It effectively lowers serum intact parathyroid hormone (iPTH) and calcium levels, thereby mitigating hypocalcemia and related gastrointestinal issues. In preclinical models of adenine-induced secondary hyperparathyroidism, Upacicalcet has demonstrated the ability to improve vascular calcification and alleviate bone disorders, while also inhibiting cortical pore formation and reducing bone fibrosis in chronic kidney disease models. This compound is valuable for investigating the mechanisms and therapeutic targets of secondary hyperparathyroidism.
