USP28 Inhibitor
USP28-IN-2 is a potent inhibitor of USP28, exhibiting an IC50 of 0.3 μM and demonstrating high selectivity for its target over other deubiquitinating enzymes such as USP2, USP7, USP8, USP9x, UCHL3, and UCHL5. This compound shows significant cytotoxic effects against cancer cells by down-regulating c-Myc levels through the ubiquitin-proteasome pathway. Additionally, USP28-IN-2 reduces ankyrase-1/2 levels in vitro and enhances the sensitivity of colorectal cancer cells to Regorafenib, making it a valuable tool for cancer research applications.
