USP28 Inhibitor
USP28-IN-4 is a potent inhibitor of Ubiquitin-specific protease 28 (USP28), exhibiting an IC50 of 0.04 μM and demonstrating high selectivity against other deubiquitinating enzymes such as USP2, USP7, USP8, USP9x, UCHL3, and UCHL5. This compound is cytotoxic to cancer cells, effectively down-regulating c-Myc levels through the ubiquitin-proteasome pathway. Additionally, USP28-IN-4 reduces ankyrin-1/2 levels in vitro and enhances the sensitivity of colorectal cancer cells to Regorafenib, making it valuable for research in cancer therapeutics and deubiquitination pathways.
