USP7 Inhibitor

USP7-IN-18 is a selective inhibitor of Ubiquitin-specific protease 7 (USP7), demonstrating an IC50 of 130.9 nM with minimal inhibition on other deubiquitinases. By binding to the catalytic domain, USP7-IN-18 effectively blocks USP7's deubiquitinase activity, leading to the degradation of oncogenic proteins such as MDM2, DNMT1, and the novel target PCLAF. This compound activates the p53-p21 pathway and demonstrates potent anti-tumor effects in colon cancer models, while also modulating the tumor immune microenvironment through both direct and immune-synergistic mechanisms.
