VX-148

IMPDH Inhibitor

VX-148 is a non-competitive inhibitor of inosine-5'-monophosphate dehydrogenase (IMPDH), targeting both IMPDH I and II with Ki values of 14 nM and 6 nM, respectively. This compound effectively suppresses the proliferation of activated human peripheral blood mononuclear cells (PBMCs) induced by T-cell mitogen (PHA) or B-cell mitogen (SPAS), demonstrating high selectivity for lymphocytes, including L1210, Jurkat T cells, and Raji B cells, while sparing non-lymphoid cells from significant toxicity. VX-148 has applications in the study of autoimmune diseases such as rheumatoid arthritis and psoriasis, as well as in organ transplantation to prevent acute rejection.
