Carboxypeptidase inhibitors target exopeptidases that trim the C-terminal residue from peptides and proteins. Most compounds here target glutamate carboxypeptidase II (GCPII), also called N-acetylated alpha-linked acidic dipeptidase (NAALADase). The enzyme cleaves the neuropeptide N-acetylaspartylglutamate (NAAG) into N-acetylaspartate and glutamate. A 1999 study reported that selective NAALADase inhibition with PMPA reduced ischemic brain injury in an animal model. The thiol-based inhibitor 2-MPPA was described in 2003 as an orally active GCPII inhibitor. ZJ 43 is a urea-based compound that inhibits both GCPII and the related glutamate carboxypeptidase III (GCPIII). The set also holds Histargin, a carboxypeptidase B inhibitor, and a carboxypeptidase G2 (CPG2) inhibitor for the bacterial folate-cleaving enzyme.
Synonyms: Carboxypeptidase inhibitor
CPA inhibitor is a potent inhibitor for carboxypeptidase A (CPA).
2-MPPA is a selective glutamate carboxypeptidase II (GCP-II) inhibitor used in the treatment of neurological disorders associated with excessive activation of glutamatergic systems. Attenuates glutamate transmission resulting in the relief of neuropathic pain.
Synonyms: CPG2 Inhibitor
Carboxypeptidase G2 (CPG2) Inhibitor is a novel Carboxypeptidase G2 (CPG2) Inhibitor, Antitumor agents.
PMPA is a potent inhibitor of the neuropeptidase N-acetylated α-linked acidic dipeptidase (NAALADase), which is a membrane-bound peptidase that hydrolyses N-acetyl-L-aspartate-L-glutamate (NAAG), a major brain peptide.
ZJ 43 is a potent inhibitor of PSM and PSMAL (glutamate carboxypeptidase II and III).
Mergetpa is a potent carboxypeptidase inhibitor that selectively blocks the conversion of kinins and B2 receptor antagonists into metabolites devoid of the C-terminal arginine. This activity plays a crucial role in modulating kinin signaling pathways, making it valuable for research in cardiovascular and inflammatory diseases. Mergetpa can serve as a tool for studying the physiological effects of kinin peptides and their potential therapeutic applications.
Histargin is a selective inhibitor of carboxypeptidase B, exhibiting an IC50 of 17 μg/ml. It demonstrates minimal inhibitory activity against carboxypeptidase A, with an IC50 greater than 100 μg/ml. This compound is useful in research applications involving protein metabolism and regulation, facilitating the study of carboxypeptidase-related processes in various biological systems. Histargin is derived from actinomycetes, adding to its relevance in natural product research.
