THX6
ClpP Activator
THX6 is a selective activator of the human ClpP protease, exhibiting an EC50 of 1.18 μM. This compound demonstrates significant cytotoxicity, particularly in ONC201-resistant cell lines with an IC50 of 0.13 μM. THX6 alters fatty acid levels, including polyunsaturated, monounsaturated, and saturated fatty acids, in diffuse intrinsic pontine glioma (DIPG) cells. Additionally, it reduces the expression of proteins associated with oxidative phosphorylation, biogenesis, and mitophagy, leading to compromised mitochondrial integrity and function, making it a valuable tool for research on DIPG.
