Antibacterial agent 327 is a potent inhibitor of Staphylococcus aureus DNA gyrase, demonstrating an IC50 of 0.28 μM against supercoiling activity. It additionally inhibits ATPase activities of both DNA gyrase and Topoisomerase IV, with IC50 values of 0.43 μM and 0.73 μM, respectively, while also affecting Topoisomerase IV decatenation activity (IC50: 2.31 μM). This compound effectively inhibits Clostridioides difficile and methicillin-resistant Staphylococcus aureus, with a minimum inhibitory concentration (MIC) of 0.78 μg/mL for both, and shows an MIC50 of 0.78 μg/mL against Escherichia coli.
