Halofuginone hydrochloride

Prolyl-tRNA Synthetase Inhibitor

Halofuginone hydrochloride is a competitive inhibitor of prolyl-tRNA synthetase, exhibiting a Ki of 18.3 nM. This compound specifically inhibits type-I collagen synthesis and mitigates osteoarthritis by suppressing TGF-β activity. Additionally, Halofuginone hydrochloride functions as a potent pulmonary vasodilator through the activation of Kv channels while blocking voltage-gated, receptor-operated, and store-operated Ca2+ channels. It has demonstrated significant anti-malaria, anti-inflammatory, anti-cancer, and anti-fibrotic properties, making it valuable for various research applications.
