CaMK

CaMK inhibitors block calcium- and calmodulin-dependent protein kinases (CaMKs) in calcium signaling. Calcium/calmodulin-dependent protein kinase II (CaMKII), a multisubunit enzyme abundant at synapses, is central to synaptic strengthening and memory formation, as reviewed in 2002. KN-93, first reported in 1991, and the related isoquinolinesulfonamide KN-62 are classical CaMKII inhibitors. The set also lists KN-93 phosphate and (E)-KN-93 phosphate, salt forms of the same inhibitor. A-484954 targets eukaryotic elongation factor 2 kinase (eEF2K), historically called CaMK III, which controls protein synthesis. The category is mixed and contains protein kinase D (PKD) inhibitors such as kb NB 142-70 and CID 755673, which act on related kinases.

    AR420626

AR420626 is a selective agonist of free fatty acid receptor 3 (FFAR3, also known as GPR41), with an IC₅₀ of 117 nM. It demonstrates anti-inflammatory, antitumor, and antidiabetic activities. AR420626 improves neurogenic diarrhea by modulating neural pathways mediated by nicotinic acetylcholine receptors (nAChRs). In cancer models, it suppresses the growth of HepG2 xenografts and inhibits hepatoma cell proliferation through apoptosis induction. Additionally, AR420626 mitigates allergic asthma and eczema and enhances glucose uptake by activating FFAR3-mediated Ca²⁺ signaling, offering potential therapeutic benefits in metabolic disorders such as diabetes.
