Gene Editing & Transfection Enhancers

Gene editing and transfection enhancers are additives that improve gene delivery and the outcome of genome editing. After a Cas9-induced double-strand break, inhibitors of DNA-dependent protein kinase (DNA-PK) such as AZD7648, NU7441 and nedisertib favor homology-directed repair (HDR). SCR7 is reported to inhibit DNA ligase IV, while RS-1 stimulates the RAD51 recombinase. Cationic polymers such as hexadimethrine bromide and protamine sulfate increase the efficiency of viral transduction. Chloroquine is added during calcium phosphate transfection and lentivirus production to reduce lysosomal degradation of plasmid DNA.

  • PolQi2
    Cat. No.
    A24142

    PolQi2

PolQi2 is a selective inhibitor of DNA polymerase theta (Polθ), specifically targeting its N-terminal helicase domain to suppress the alternative end-joining (alt-EJ) DNA repair pathway. By inhibiting Polθ, PolQi2 enhances the precision and integration efficiency of CRISPR/Cas9-mediated gene editing across diverse genomic loci and cell lines. Furthermore, when used in combination with DNA-PK inhibitors, PolQi2 significantly reduces off-target effects associated with Cas9 activity. These properties make PolQi2 a valuable tool for improving the fidelity and efficiency of gene editing applications in both basic research and potential therapeutic contexts.

  • PolQi1
    Cat. No.
    A37668

    PolQi1

PolQi1 is a selective inhibitor of the Polθ domain of DNA polymerase, primarily targeting the Polθ-mediated microhomology end joining (TMEJ/alt-EJ) pathway. By inhibiting this pathway, PolQi1 significantly reduces insertion/deletion events and imprecise editing during DNA repair processes. This reagent enhances the efficiency and accuracy of homology-directed repair (HDR) and Prime editing, minimizing off-target effects. In combination with DNA-PK inhibitor AZD-7648, PolQi1 provides effective dual pathway regulation, making it a valuable tool for gene editing research, particularly in optimizing systems like CRISPR-Cas9 for challenging cell types such as primary hepatocytes and mouse embryos.
